The Lake Rune increases your physical defense. The vast majority of enemy attacks in the game deal physical damage, so this is very useful. Stacking physical damage reduction can help a lot if you like to rely on the Regain system to regenerate your health, too. Just be extra careful of enemies with special attacks and those that buff their weapons with elemental damage; physical defense won't help you against them.
