The Communion Runes increase the number of Blood Vials you can hold. This is very helpful, particularly if you have difficulty making use of the Regain system. By default, each of your Blood Vials heals you for about 40% of your maximum health, but this effect can be increased using the Radiance Oath Rune; Communion and Radiance work very well together. If you often use Blood Bullets or simply don't like Regain, this can help. Having the extra vials can also be very useful to keep yourself topped-off if you like to rely on the Fool gem effect. On the other hand, if you keep the Pulsing effect on your weapons this Rune may be unnecessary.
